IL-2对COPD患者CD8~+ T细胞频率及功能的影响
Effect of IL-2 on frequency and function of CD8~+ T cells in COPD patients
【摘要】 目的探讨IL-2在慢性阻塞性肺疾病(COPD)中的免疫调节作用。方法采集45例COPD急性发作期患者、33例COPD缓解期患者以及25例健康人外周血,采用ELISA检测血清中IFN-γI、L-2浓度。提取PBMC后分别设置IL-2刺激组和未刺激组,多色流式分析技术检测CD8+T细胞频率,胞内细胞因子染色技术检测CD8+IFN-γ+T细胞的频率。结果 COPD患者急性发作期IL-2I、FN-γ浓度较临床缓解期和对照组显著增高(P均<0.01);与未刺激组相比I,L-2刺激组CD8+T细胞频率及分泌IFN-γ显著增加(P<0.05)。结论 IL-2可通过对CD8+T细胞的免疫调节效应参与COPD的发生发展过程。
【Abstract】 Objective To investigate the immunoregulatory role of IL-2 in the chronic obstructive pulmonary disease(COPD).Methods IL-2 and IFN-γ in serum from 78 patients with COPD and 25 healthy subjects were quantitively analyzed using enzyme-linked immunosorbent assay(ELISA).The frequency of CD8+ T cells was analyzed by flow cytometry.IFN-γ was analyzed using intracellular cytokine straining.Results The serum concentrations of IL-2 and IFN-γ in acute exacerbations of COPD patients were significantly higher than those in stable and control groups(all P<0.01).There were higher frequencies of circulating CD8+T cells and CD8+IFN-γ+T cells in the IL-2 stimulation group than those of no IL-2 stimulation group(all P<0.05).Conclusons IL-2 play an important role in the genesis and development of COPD by immunoregulating the frequency and function of CD8+ T cells.
