帕金森病患者时钟基因启动子区甲基化分析

【摘要】 目的分析帕金森病(Parkinson’s disease,PD)患者和正常老年对照组时钟基因启动子区甲基化水平,探讨帕金森病患者时钟基因异常表达的机制。方法采用甲基化特异性PCR(MSP)法,检测206例帕金森病患者和181例正常老年对照者的7种时钟基因启动子甲基化水平。结果 7种时钟基因中,cry1和npas2启动子区存在明显甲基化。另5种时钟基因启动子(per1、per2、cry2、clock、bmal1)未发现明显甲基化。PD患者中npas2甲基化频率明显下降。结论 PD患者时钟基因的异常表达可能与时钟基因启动子区甲基化的改变有关。

【Abstract】 Objective To study promoter methylation of seven clock genes in Parkinson’s disease(PD) and determine whether abnormal promoter methylation contributes to the dysregulated expression of clock genes. Methods Methylation frequencies of seven clock promoters were analyzed in 206 PD patients and 181 healthy controls using methylation-specific PCR(MSP) method. Results Most clock gene promoters were devoid of methylation.Methylation was only detectable in the cry1 and npas2 promoters.Of note,the methylation frequency of the npas2 promoter was significantly decreased in PD patients. Conclusion These results suggest that altered promoter methylation may contribute to the abnormal expression of clock genes in PD.
