毛细管电泳电化学法检测大鼠腹腔肥大细胞提取液中的抗坏血酸
Determination of ascorbic acid in rat peritoneal mast cells extract by capillary electrophoresis with electrochemical detection
【摘要】 利用毛细管电泳电化学法检测了大鼠腹腔肥大细胞提取液中的抗坏血酸,考察了缓冲液的pH值、缓冲液浓度、分离电压、检测电势等实验参数对检测抗坏血酸的影响.在最佳实验条件下,检测抗坏血酸的浓度范围是5.0×10-6~5.0×10-4mol.L-1,线性回归系数为0.9962,浓度检测限(S/N=3)为1.7×10-6mol.L-1.以此方法检测大鼠腹腔肥大细胞提取液中的抗坏血酸的含量为2.1×10-6mol.L-1,样品的加标回收率为93~97%.
【Abstract】 Ascorbic acid in rat peritoneal mast cells extract was detected by capillary electrophoresis with electrochemical detection.The effects of the running buffer pH,running buffer concentration,separation voltage and detecting potential on the measurement were studied.Under the optimum conditions of detection,the linear range of ascorbic acid was 5.0×10-6 ~5.0×10-4 mol·L-1.The correlation coefficient of the method was 0.9962.The limit of detection(S/N=3) was 1.7×10-6 mol·L-1.Ascorbic acid in rat peritoneal mast cell extract was 2.1×10-6 mol·L-1.The recoveries were 93%~97%.
