【摘要】 目的:探讨祛瘀化痰、散结消癥中药复方对肾小球系膜细胞增殖及ECM合成调控系统的影响。方法:以脂多糖为刺激因子,作用于体外培养的大鼠肾小球系膜细胞,设立空白对照组、川芎组、川芎+鳖甲组、川芎+海藻组、川芎+鳖甲+海藻组(高、中、低剂量组),以MTT比色法观察各组肾小球系膜细胞增殖情况,ELISA法观测细胞上清中FN水平的变化。结果:川芎组、川芎+鳖甲组、川芎+鳖甲+海藻(高、中剂量)组均能不同程度地抑制肾小球系膜细胞增殖,减少FN的含量,中以川芎+鳖甲+海藻组(高剂量)作用最明显,且随着用药浓度增加,作用增强。结论:祛瘀化痰、散结消癥中药复方具有干预肾小球硬化的作用,而这一作用可能与有效抑制系膜细胞增殖、下调系膜细胞上清中FN水平,减少ECM合成有关。更多还原

【Abstract】 Objective:To investigate the effect of the herbal compound prescriptions of removing blood stasis and resolving phlegm,dispersing abdominal stagnated mass on proliferation of glomerular mesangial cells and extracellular matrix(ECM) synthesis.Methods:After stimulated by LPS,in-vitro cultured glomerular mesangial cells were divided into blank control group.Rhozoma Chuanxiong(Chuanxiong) group,Chuanxiong+Carapax Trionycis(Biejia) group,Chuanxiong+Sargassum(Haizao) group,and high-,middle-and low-dosage Chuanxiong+Biejia+Haizao groups.MTT colorimetric assay was used to observe the proliferation of glomerular mesangial cells of each group and fibronectin(FN) level in the cultured supernatant was detected by ELISA.Results:Proliferation of glomerular mesangial cells was inhibited,and FN level was decreased in Chuanxiong group,Chuanxiong+Biejia group,and high-,middle-and low-dosage Chuanxiong+Biejia+Haizao groups to various degrees,and the effect was most obvious in high-dosage Chuanxiong+Biejia+Haizao group,the effect being in concentration dependent manner.Conclusion:The herbal compound prescriptions of removing blood stasis and resolving phlegm,dispersing abdominal stagnated mass has interference on glomerular sclerosis,and its mechanism may be related to the inhibition of mesangial cells proliferation,down-regualtion of FN level in supernatants liquid of mesangial cells and decrease of ECM synthesis.更多还原
