长期能量限制对成年雌性大鼠生殖寿命的影响
Effect of long-term calorie restriction on the reproduction of female rats
【摘要】 目的探讨长期能量限制对成年雌性大鼠生殖寿命的影响。方法 6月龄雌性SD大鼠30只,分为正常饮食组、能量限制(CR)20%组和CR40%组,记录正常饮食(NC)组每日摄食量,并将此摄食量的80%和60%分别喂养CR20%组和CR40%组大鼠,监测大鼠体重及动情周期变化,12个月后取大鼠卵巢。卵巢组织切片由苏木精-伊红染色,观察卵泡储备数量和不同发育阶段的卵泡比例。结果 NC组体重持续上升,而CR20%组体重小幅增加,CR40%组体重持续下降。CR40%组大鼠的卵泡储备数量(105.7±9.5)和原始卵泡数(55.0±6.8)与NC组(51.2±5.4,18.3±3.7)相比差异均有统计学意义(P<0.001,P<0.001),原始卵泡比例(52.1%±2.9%)与对照组(35.8%±5.5%)相比差异有统计学意义(P<0.001),而成熟卵泡比例(6.8%±1.9%)和发育卵泡比例(41.2%±3.0%)与NC组(15.6%±6.2%,48.9%±4.2%)相比差异亦有统计学意义(P<0.001,P<0.05)。CR20%组卵泡储备数量和不同发育阶段的卵泡比例与NC组相比差异无统计学意义。结论长期能量限制可能通过抑制卵泡发育启动、减少发育卵泡和成熟卵泡的数量,从而减少卵泡的消耗,有益于生殖寿命延长。
【Abstract】 Objective To investigate the effects of Long-term calorie restriction on the extension of ovarian reproductive life in female SD rats.Methods 30 mature female SD rats were divided into three groups:normal control(NC)(fed standard rodent chow ad libitum),20% calorie restriction(CR20%) group(fed with 80% food of NC group),and 40% calorie restriction(CR40%) group(fed withn 60% food of NC group).The body mass was monitored monthly,and the estrous cycle patterns were determined by vaginal smears every two months.Ovaries were collected after 12 months treatment.Ovarian tissue sections were prepared and stained by hematoxylin and eosin.The total number of ovarian follicles and the ratio of follicles at different developmental stages were counted and calculated.Results 1) Body mass:There was a significant increase in the body mass of rats from the NC group.There was no significant increase in the CR20% rats.However,the body mass of CR40% rats showed a fluctuant decline.2) Ovarian follicle development:There was a significant increase in the total number of primordial follicles(P<0.001) and healthy follicles(P<0.001) in CR40% rats.The percentage of primordial follicles(P<0.001) in CR40% rats was significantly higher than those of NC rats.However,the percentages of developmental follicles(P<0.05) and mature follicles(P<0.001) in CR40% rats were significantly lower than the NC rats.Rats in the CR20% group had a comparable total number and ratio of follicles to the control group.Conclusion Long-term calorie restriction may inhibit the transition of primordial to developing follicles and decrease the number of developing follicles and mature follicles.These results suggest that CR can inhibit the reproductive axis,increase ovarian follicular reserves,and prolong the reproductive life of ovary.
