磷脂酶A2活性与肺间质纤维化形成相关性研究

【摘要】 目的研究磷脂酶A(2PLA2)活性及其相关炎症介质在肺间质纤维化形成中的作用。方法 64只3月龄健康SD大鼠,随机分为正常对照组(A组),肺纤维化模型组(B组),每组于给药后第3、7、14、28d各随机处死8只,光镜下观察肺组织形态学变化、测定肺组织匀浆中羟脯氨酸(HyP)含量、PLA2活性及支气管肺泡灌洗液(BALF)中前列腺素E2(PGE2)、白三烯C4(LTC4)和转化生长因子-β1(TGF-β1)含量变化。结果第3、7、14、28d,模型组肺组织匀浆中PLA2活性和BALF中的PGE2、LTC4及TGF-β1含量均较正常对照组显著增高(P<0.01),肺组织HyP含量于第7d开始较正常对照组增高(P<0.05),第28d达最高峰。相关性分析显示,PLA2与LTC4、PGE2,TGF-β1及HyP之间均存在正相关性;LTC4、PGE2及TGF-β1三者之间也有一定的相关性。结论 PLA2可能通过LTC4、PGE2、TGF-β1在体内综合作用结果参与了肺间质纤维化的发生发展过程。

【Abstract】 Aim To study the relationship among the phospholipase A2(PLA2)activity,its metabolic products,and pulmonary interstitial fibrosis.Methods There 64 healthy Sprague-Dawley rats aged three months,were randomly divided into Group A,the normal control group and Group B,the model group each consisted of 32 cases treated with bleomycin.At day 3,7,14 and 28 after injection of bleomycin,eight rats in each group were randomly chosen and killed.The right lungs of dead rats were removed and appropriately processed for hematoxylin and eosin(H&E)stain,histologically observed under light microscope.The hydroxyproline content and the PLA2 activity in pulmonary homogenate were measured with alkaline hydrolysis assay and acid modified microtitrimetic method.The levels of leukotriene C4(LTC4)、prostaglandin E2(PGE2)、transforming growth factor-β(1TGF-β1)in bronchoalveolar lavage fluid(BALF)were measured with enzyme-linked immunosorbent assay(ELISA).Results At day 3,7,14 and28 after intratracheal instillation of bleomycin in rats of group B,the PLA2 activity in lung homogenate and the levels of its metabolic products PGE2,LTC4 as well as TGF-β1 in BALF increased significantly compared with those in group A(P<0.01).And lung hydroxyproline concentration began to increase markedly at day 7 compared with those in group A(P<0.05),reaching its maximum at day 28.PLA2 was positively correlative with PGE2,LTC4,TGF-β1 and hydroxyproline.Additionally,there existed correlation among PGE2,LTC4 and TGF-β1.Conclusion PLA2 play an important role in the formation of pulmonary interstitial fibrosis probably due to comprehensive effect of LTC4,PGE2 and TGF-β1 in vivo.
