大肠锯齿状腺瘤和腺癌组织HIF-1α表达及其与VEGF和血管密度相关性的探讨

【摘要】 目的:探讨大肠锯齿状腺瘤和腺癌组织HIF-1α的表达及其与VEGF和血管密度的关系。方法:采用免疫组化法检测HIF-1α和VEGF蛋白的表达。用CD34单克隆抗体标记血管内皮细胞并计数。结果:锯齿状腺瘤HIF-1α蛋白阳性率(62.5%)显著高于正常大肠黏膜组(13.3%),χ2=15.78,P<0.01;显著低于大肠癌组(85.0%),χ2=4.8,P=0.028。锯齿状腺瘤VEGF蛋白阳性率(53.1%)显著高于正常大肠黏膜组(10.0%),χ2=13.17,P<0.01;而与大肠癌组(72.5%)比较差异无统计学意义,χ2=2.89,P=0.089。HIF-1α表达与VEGF正相关(r=0.317,P=0.041);与MVD正相关,r=0.468,P=0.02。结论:HIF-1α促进肿瘤血管生成,参与大肠锯齿状腺瘤癌变。

【Abstract】 OBJECTIVE:To explore the relationship of hypoxia-inducible factor alpha protein expression with vascular endothelial growth factor and microvessel density in colorectal serrated adenoma and colorectal cancer.METHODS: HIF-1α and VEGF protein expression were determined by immunohistochemisty in colorectal serrated adenoma and colorectal cancer.Microvessel density was determined by anti-CD34 immunostaining.RESULTS: Positive expression rate of HIF-1α protein in colorectal serrated adenoma group was significantly higher than that in normal group(62.5% vs 13.3%,χ2=15.78,P<0.01),but positive expression rate of HIF-1α protein in colorectal serrated adenoma group was significantly lower than that in colorectal cancer.(62.5% vs 85.0%,χ2=4.8,P=0.028).Positive expression rate of VEGF protein in colorectal serrated adenoma gruop was significantly higher than that in normal group(53.1% vs 10.0%,χ2=13.17,P<0.01) and was no difference in colorectal cancer(53.13% vs 72.5%,χ2=2.89,P=0.089).The expression of HIF-1α protein was positively correlated with the VEGF protein expression and MVD.(r=0.317,P=0.041;r=0.468,P=0.02).CONCLUSION: Tumor angiogenesis is induced by HIF-1α,and HIF-1α involve in the transforming from colorectal serrated adenoma to colorectal cancer.
