【摘要】 目的探讨慢病毒介导的Clusterin(CLU)基因沉默的RNA干扰效果及其对膀胱癌EJ细胞增殖和转移的影响。方法构建靶向CLU基因的慢病毒干扰载体,用Westernblot检测其对膀胱尿路上皮癌细胞株EJ的干扰效果,用MTT、细胞划痕实验、Transwell侵袭实验等方法分别检测CLU沉默后膀胱癌EJ细胞在增殖、侵袭和转移等方面的细胞生物学改变。结果靶向CLU基因的慢病毒干扰载体感染膀胱癌EJ细胞后,CLU基因在蛋白水平的表达明显下降。EJ细胞的增殖、侵袭和转移能力均受到抑制。结论 CLU慢病毒干扰载体可有效沉默膀胱癌EJ细胞内源性CLU基因,抑制膀胱癌EJ细胞的增殖、侵袭和转移。更多还原

【Abstract】 Objective To testify the silence efficiency of lentivirus mediated RNA interference targeting CLU and the influence to the proliferation and metastasis of bladder cancer EJ cells.Methods Lentivirus mediated RNA interference vector targeting CLU was constructed.Western blot was recruited to testify the silence efficiency.After that,MTT,wound healing assay,Transwell invasion assay were applied to examine the effect of CLU silence on the proliferation,invasion and in EJ cells.Results After bladder cancer EJ cells were infected by lentivirus mediated RNA interference vectors targeting CLU,expression of CLU gene decreased significantly.The proliferation,invasiveness and metastasis of EJ cell line was suppressed.Conclusions Lentivirus mediated RNA interference targeting CLU can effectively silence endogenous CLU expression in bladder cancer EJ cells,and inhibit proliferation,invasiveness and metastasis in EJ cells.更多还原
