新的肿瘤转移抑制基因LASS2在不同侵袭潜能的膀胱癌细胞中表达的体外研究
Expression of a new tumor metastasis supressor gene LASS in human bladder cancer cell lines with different invasion ability
【摘要】 目的探讨新发现的肿瘤转移抑制基因人源性长寿保障基因Ⅱ型(homo sapiens longevity assurance homologue2,LASS2)与人膀胱癌细胞侵袭潜能的关系。方法通过细胞生长曲线和体外侵袭实验,确定膀胱癌BIU-87、EJ、T24和EJ-M3细胞的增殖和侵袭能力;运用实时定量PCR法和Western blot法检测4株细胞中LASS2基因的表达。结果 4株膀胱癌细胞中,EJ-M3的增殖和侵袭能力最强;增殖和侵袭能力高的细胞系EJ-M3和BIU-87的LASS2mRNA表达降低(P<0.001),而在低转移能力的人膀胱癌细胞株T24和EJ细胞中则相对较高表达。结论 LASS2基因表达的降低,可能与膀胱癌细胞的增殖和侵袭有关。
【Abstract】 Objective To investigate expression and potential role of a new tumor metastasis supressor gene LASS2 in human bladder cancer.Methods Reproductive activity and invasion ablity of BIU-87,EJ,T24 and EJ-M3 bladder cancer cell lines were identified by using the methods of auxodrome and in vitro invasion assay.The expression of LASS2 gene in BIU-87,EJ,T24 and EJ-M3 cell lines was examined using real-time quantitative PCR and Western blot.Results The most aggressive human cancer cell line was EJ-M3 among the four cell lines.The expression of LASS2 mRNA was significantly related with diverse proliferation and invasion.At the protein level,we found that the more aggressive of cancer cell line,the less LASS2 protein expression.Conclusions LASS2 expression may be correlated with the development and progression of human bladder cancer and may be a prognostic indicator for this cancer.
