抗凋亡蛋白Livin在急性白血病中的表达

【摘要】 目的研究Livin在急性白血病中的表达,探讨其临床意义。方法采用半定量逆转录—聚合酶链反应(RT-PCR)检测40例急性白血病(AL)患者(AL组)LivinmRNA的表达水平,其中初治患者16例(初治亚组)、复发患者11例(复发亚组)、完全缓解患者13例(缓解亚组)。另选10例健康人(对照组)的标本作正常对照。结果初治亚组中LivinmRNA的表达较对照组明显升高(P<0.05),治疗缓解后的表达水平下降(P<0.05),复发时又再次升高;初治亚组Livin阳性的患者缓解率(14.3%)低于表达阴性的患者(77.8%)(P<0.05)。LivinmRNA的表达与AL患者性别、年龄无关(P>0.05)。结论 Livin基因的过度表达参与了AL的发病,并且是AL复发的高危因素。Livin基因的高表达是判断AL预后不良的指标之一。

【Abstract】 Objective To research the expression of Livin in acute leukemia,and explore its clinical significance.Methods The expression of Livin mRNA of 40 cases patients with acute leukemia(AL) was measured by semi-quantity reverse transcription polymers chain reaction(RT-PCR).In which,16 cases newly diagnosed patients(newly diagnosed sub-group),11 cases patients relapsed(recurrence sub-group),13 cases patients with complete remission(remission sub-group).Another 10 healthy people were as control group.Results The expression of Livin in newly diagnosed sub-group was higher than that in control group(P<0.05),while it decreased in patients with remission sub-group(P<0.05);In relapsed patients,the expression of Livin increased again.The response rate of patients with Livin positive(14.3%) was lower than that of patients with Livin negative(77.8%) in newly diagnosed sub-group(P<0.05).The expression of Livin mRNA in patients with AL had nothing to do with sex and age(P>0.05).Conclusion Overexpression of Livin may play a synergic role in the pathogenesis of leukemia and may be a high risk factor for relapse in AL.High expression of Livin can serve as one of the markers of poor prognosis in AL.
