环介导等温扩增技术检测乳粉中肺炎克雷伯菌
Development of Loop-mediated Isothermal Amplification(LAMP) Detection of Klebsiella pneumoniae in Milk Powder
【摘要】 以荚膜脂多糖(capsular polysaccharide,CPS)合成调控子resA为靶基因片段,设计了肺炎克雷伯菌特异性引物,建立了环介导恒温扩增技术(Loop-Mediated lsothermal Amplification,LAMP)检测方法.结果表明,3对LAMP引物对肺炎克雷伯菌不仅有菌种专一性,而且,具有相当高的灵敏度(2 CFU/100 g).因此,该方法有利于更快更准确的鉴定食品中肺炎克雷伯氏菌.
【Abstract】 Klebsiella pneumoniae(K.pneumoniae)is a Gram-negative bacteria found in the normal flora of the mouth,skin,and intestines,which was recognized as a causative agent of pneumonia,meningitis,and blood poisoning ere food born diseases.In present study,3 pairs of primers targeted on the regulator of capsule transcription A gene(RcsA)of K.pneumonia were designed and used the method for loop-mediated isothermal amplification(LAMP)assay. We found that with our K.pneumoniae specific LAMP primers could efficiently detected the rcsA gene within I hour at a minimal amount of bacteria(about 2 CFU/a reaction).
