全反式维甲酸对裸鼠子宫内膜癌治疗作用的实验研究
Effects of ATRA on endometrial cancer in nude mice: A experimental study
【摘要】 目的:探讨全反式维甲酸(ATRA)对裸鼠子宫内膜癌的作用及其机制。方法:建立子宫内膜癌裸鼠模型,随机分为5组:ATRA低剂量组(建模第11天予ATRA 20 mg.kg-1.d-1,共20 d)、ATRA高剂量组(建模第11天予ATRA 40 mg.kg-1.d-1,共20 d)、顺铂组(建模第11天予顺铂3 mg.kg-1.d-1,共20 d)、ATRA对照组(建模同时予ATRA 20 mg.kg-1.d-1,每周5次,共4周)、空白对照组(每天灌入食油0.5 ml,共30 d)。治疗期间测量皮下癌瘤大小并绘制生长曲线,观察肿瘤生长抑制率,并用RT-PCR法检测癌组织中肝细胞生长因子c-met的表达。结果:肿瘤生长抑制率ATRA高剂量组(67.34%)明显高于ATRA低剂量组(41.18%)(P<0.05),而与顺铂组(68.06%)相当(P>0.05);ATRA高剂量组与顺铂组死亡率比较无明显差异(P>0.05)。各组裸鼠体重实验前后比较无显著性差异(P>0.05)。裸鼠癌瘤组织中可检测到c-met的表达,ATRA能抑制c-met的表达,ATRA低剂量组、ATRA高剂量组及顺铂组与空白对照组比较差异有统计学意义(P<0.05),ATRA低剂量组、ATRA高剂量组、ATRA对照组之间c-met表达差异无统计学意义(P>0.05)。结论:ATRA能够通过抑制c-met的表达抑制HGF/c-met介导的子宫内膜癌增殖,有希望成为治疗子宫内膜癌的药物之一。但ATRA并不能阻止移植瘤的形成,有必要采用联合用药以提高其抑瘤作用。
【Abstract】 Objective: To investigate effects of all transretinoic(ATRA)on endometrial carcinoma in nude mice and its mechanism.Methods: Nude mice with transplanted HEC-1 tumor were randomly divided into 5 groups: ATRA low-dose group(ATRA 20 mg·kg-1·d-1,for a total 20 ds,from the 11th day animal model was made),high-dose group(ATRA 40 mg·kg-1·d-1,for a total 20 days,from the 11th day animal model was made),cisplatin group(3 mg·kg-1·d-1, for a total 20 days from the 11th day animal model was made),ATRA control group(ATRA 20 mg·kg-1·d-1 from the day we began to make animal model.5 times a week, for a total 4 weeks),blank control group(oil 0.5ml·d-1,for a total 30 days). The tumor size was measured and the growth curve of the tumors was drew.The growth inhibiting rate of the tumor in nude mice was observed and the expression of the hepatocyte growth factor c-met was detected by the method of RT-PCR.Results: Tumor growth inhibiting rate of high-dose group(67.34%) was significantly higher than that of low-dose group(41.18%)(P<0.05),but was similar with cisplatin group(68.06%)(P>0.05).The mortality of high dose group was similar with cisplatin group too(P>0.05).There was no difference in each group mice weight before and after experiment(P>0.05).The hepatocyte growth factor receptor c-met was detected in tissue of endometrial cancer of nude mice.ATRA could inhibite expression of c-met.There were significantly different in expression of c-met comparing low-dose group,high-dose group and cisplatin group with control group.There were no significanty diffference among three different retinoic acid groups.Conclusions: ATRA can inhibit the growth of tumor,which may be through the inhibiting the HGF/c-met path,ATRA may be a potential drug to treat the endometrial cancer.However,ATRA can not prevent tumor formation,it is necessary to increase their use of combination.
