H2-K/β2-m双转基因小鼠的制备及鉴定

【摘要】 为了深入地研究MHCⅠ类分子在神经元活动中的作用,作者利用免疫荧光技术在胎鼠原代神经元水平证实了H2-K在皮层神经元的表达,从C57 BL/6j小鼠脾脏中提取RNA,逆转录并进行PCR扩增获得小鼠H2-K和β2-m基因,并分别克隆入载体pBluescripte-Fzd 9-Promoter中,然后双酶切获得Fzd 9-Promoter-H2-K和Fzd 9-Promoter-β2-m。将这两种基因同时显微注射至小鼠受精卵中,制作双转基因小鼠,得到了两只单转β2-m和3只H2-K/β2-m共转的转基因小鼠,为下一步的工作奠定了基础。

【Abstract】 In order to study the function of MHCⅠmolecular in mouse neuron,H2-K and β2-m cDNAs were amplified from the mouse(strain C57BL/6j) spleen RNA by using RT-PCR,and cloned into the pBluescripte-Fzd9-Promoter vector respectively.After cleaved the Fzd9-Promoter-H2-K and Fzd9-Promoter-β2-m fragments out of the recombinant plasmids by double restriction enzyme digestion,these two fragments were mixed together and injected into the fertilized mouse egg to get the double transgenic mouse.Three strains of H2-K/β2-m double transgenic mice and two strains of β2-m transgenic mice were generated and ready for the further studying of the role of MHC in nerve system.
