鹅胚脾淋巴细胞选择机制研究

【摘要】 取发育到第14天的鹅胚胎(E14)到1日龄雏鹅的脾脏用免疫荧光双标法研究CD4和CD8抗原表达,用Hoeschst33342的荧光染色方法分析细胞凋亡率,探索鹅胚脾淋巴细胞选择机制。结果显示:E14脾初具轮廓,E19脾出现红髓和白髓,1日龄时脾被膜增厚,脾小结数目增多,脾结构完整。CD4和CD8共表达于脾的红髓和被膜上,白髓中表达量较少。鹅胚脾的CD4+CD8+表达量为1%~30%;1日龄雏鹅的双阳性面积百分比与胚胎鹅E14、E19、E24、E27差异极显著(P<0.01);1日龄雏鹅的凋亡率与胚胎鹅E14、E24差异极显著(P<0.01)。表明鹅胚脾的CD4+CD8+表达与细胞凋亡存在协同关系。

【Abstract】 The selection mechanism of lymphocytes of the fetal spleen at the age from E14(14-day embryos)to 1 day-old(1 d after hatching)were studied by morphological observation,and the expression of CD4 and CD8 was studied using the method of double-labeled immunofluorescence,and the apoptosis was also analyzed with the fluorescent dye Hoeschst 33342.The results were shown as follows:the spleen formed until the spleen at E14.The red and white pulp appeared at E19.The spleen capsule became thicker at 1 day-old.The cell number in splenic corpuscle increased,and the spleen structure grew mature.CD4+CD8+ immunofluorescence was distributed in the splenic red pulp and the membrane,but absent in white pulp.In goose embryo,the expression of CD4+CD8+ was around from 1% to 30%.Compared with 1 day-old,the expression of CD4+CD8+ of E14,E19,E24 and E27 was significantly different(P<0.01);Compared with 1 day-old,the apoptotic rate of E14 and E24 was significantly different(P<0.01).It was suggested that there was a coordination relationship between expression of CD4+CD8+ and apoptosis in the spleen of the goose.
