β-catenin在IL-24抑制肝癌细胞的血管生成和肿瘤生长中的作用

【摘要】 目的研究IL-24对肝癌细胞在鸡胚绒毛尿囊膜(chick chorioallantoic membrane,CAM)上的血管生成和肿瘤生长的影响及其机制的初探。方法实验采用病毒感染复数(multiplicity of infection,MOI)为5的重组慢病毒IL-24和慢病毒空载体Mock分别感染肝癌细胞HepG2,24 h后接种于CAM,4 d后观察CAM上形成的肿瘤背面血管数目,并剥离肿瘤组织称其重量,继而进行Western blot检测肿瘤组织中β-catenin的表达。结果 CAM实验显示,与Mock感染的HepG2细胞相比,慢病毒IL-24感染的HepG2细胞形成的肿瘤背面血管分支数目和肿瘤重量明显下降。Western blot结果发现慢病毒IL-24感染的HepG2细胞形成的肿瘤组织中β-catenin显著低于Mock组。结论 IL-24能够抑制肝癌细胞的肿瘤血管生成和肿瘤生长,这种作用可能是通过下调β-catenin的表达来实现的。

【Abstract】 Interleukin-24(IL-24),a cytokine belonging to the IL-10 family,selectively induces apoptosis in cancer cells without harming normal cells by modulating many signal pathways,but the precise molecular mechanism by which IL-24 inhibits hepatocarcinoma cell-induced angiogenesis and tumor growth requires further clarification.Here,we used the chick chorioallantoic membrane(CAM) model to investigate the role of β-catenin signaling in IL-24-induced inhibition of tumor angiogenesis and growth.The results showed that the blood vessel branches and weight of tumors derived from HepG2 cells transduced with lentiviral IL-24 decreased significantly compared with those of transduction with Mock.And Western blot assay indicated the expression of β-catenin in tumor tissues with lentiviral IL-24 dramatically down-regulated compared with those of transduction with Mock.These data suggest that IL-24 can down-regulate the β-catenin signaling,which contributes to suppress angiogenesis and tumor growth of human hepatocarcinoma cells.
