膀胱肿瘤蛋白质表达谱及生物标记发现

【摘要】 目的分析浅表膀胱肿瘤蛋白质表达谱的特点及膀胱肿瘤-间质相互作用,探讨膀胱癌尿液蛋白质芯片的发现。方法前瞻性收集浅表膀胱移行细胞癌标本4例,激光捕获显微切割获得纯化的浅表膀胱肿瘤细胞以及间质细胞;二维液相色谱分离多肽混合物,电喷雾串联质谱鉴定蛋白质;生物信息学方法分析鉴定蛋白质。所得结果与已发表文献进行比对。结果 4例浅表膀胱移行细胞癌标本中775个蛋白质共同表达。612个蛋白质具有基因本体论(GO)生物学途径注解。40、25个GO生物学途径术语为浓集、缺失表达。表达谱浓集表达蛋白质集中于代谢途径。117个蛋白质共同表达于肿瘤细胞以及间质细胞,其中73个定位于已知生物通路。属于细胞粘附、细胞增殖、细胞分化类别的蛋白质是尿液中检测肿瘤的良好的生物标记候选蛋白。结论浅表膀胱肿瘤蛋白质表达谱研究对理解肿瘤细胞生物学、肿瘤间质相互作用提供了理论基础,并为从尿液中发现生物标记提供了候选蛋白。

【Abstract】 Objective To sumcnarize the expression profile of transitional cell carcinoma by shotgun proteome strategy,and based on which the tumor-stroma interaction and biomarker panel discovery from urine were discussed.Methods Specimens of 4 bladder transitional cell carcinoma were prespectively collected.Cancer cells and stroma were purified by laser capture microdissection(LCM).Peptide mixture was separated by two-dimensional liquid chromatography(2D-LC) followed by electrospray ionization-tandem mass spectrometry(ESI-MS/MS) identification.And then bioinformatics of the identified proteins were analyzed and compared with the published literature.Results A total of 775 proteins commonly expressed in the 4 specimens,612 of which had biological process annotation as to the Gene Ontology(GO).There were 40/25 GO terms exhibited as enriched/depleted within the biological process annotation.The enriched GO terms centered on metabolism process.117 proteins expressed both in cancer cells and stroma.Among them,73 proteins were located in well-known pathways.Proteins belonging to the term of cell adhesion,cell proliferation,cell differentiation were good candidate biomarkers for cancer detection from urine.Conclusions The present expression profile provids valuable information for the understanding of cell biology,tumor-stroma interaction as well as the discovery of biomarkers from urine.
