高效毛细管电泳分离和测定丹红注射液中水溶性活性成分

【摘要】 目的探讨高效毛细管电泳分离和测定丹红注射液中水溶性活性成分。方法磷酸盐缓冲溶液浓度为50 mmol/L,pH 7.2,电压为15 kV,温度为25℃,检测波长为210 nm,未涂层石英毛细管(内径为75μm),毛细管总长为48.5 cm,有效柱长为40 cm,在优化的电泳分离条件下分析得到丹红注射液中水溶性活性成分原儿茶醛、丹参素、原儿茶酸的电泳图,考察3种分析物的线性、检测限和方法的重现性。结果丹红注射液中的原儿茶醛、丹参素和原儿茶酸混合物在11 min内完全基线分离。结论高效毛细管电泳可用于丹红注射液的定量分析。

【Abstract】 Objective To explore the separation and determination of water-soluble active components in Danhong injection by high performance capillary electrophoresis analysis technique. Methods 50 mmol/L phosphate buffer,pH 7.2,working voltage at 15 kV,temperature at 25℃were used.Capillary electrophoresis was performed using a uncoated fused-silica capillary whose length is 48.5 cm and effective column length is 40 cm.Diode array detection was set at 210 nm. Water-soluble active ingredients,procatechuic aldehyde,Danshensu and procatechuic acid in Danhong injection electrophoresis were analyzed under the optimization of the electrophoretic separation conditions.The linear of those three compounds,detection limits and reproducibility of the method were studied.Results Under the optimizing conditions,procatechuic aldehyde, Danshensu and procatechuic acid in Danhong injection could be successfully determined within 11 min.Conclusion High performance capillary electrophoresis method is suitable for quantitative analysis of Danhong injection.
