阳离子乳剂介导的基因载体给药
Cationic Emulsions Mediated Gene Carrier Administration
【摘要】 目的利用阳离子乳剂包裹用于治疗的基因片段,通过转染机体细胞达到基因治疗的目的。方法取DOTAP溶解于氯仿,真空干燥去除有机乳剂后,用超声细胞破碎仪在冰水中震荡后加入角鲨烯继续超声即得到阳离子乳剂,细胞转染实验之前,把乳剂与质粒DNA混合均匀,室温静置即得到带正电阳离子乳剂-DNA复合物。结果阳离子乳剂包裹的基因片段能成功转染机体细胞。结论包裹基因载体的阳离子乳剂表面携带正电荷与机体细胞膜表面的负电荷吸引并结合,有效地增加了细胞转染的成功率,是一种方便、经济、实用的基因给药新方法。
【Abstract】 Objective To cure disease,the curative gene fragment in the cationic emulsions was prepared. Methods DOTAP was dissolved in chloroform which was removed by vacuum drying.The cationic emulsions were prepared by shocking the mixture,which contained DOTAP and spinacene,and then added plasmid.the cationic emulsions-DNA compounds were prepared before cell transfection experiment. Results The curative gene fragment in the cationic emulsions could transfect cells successfully. Conclusion The curative gene fragment in the cationic emulsions,which attract and combine the negative charge in the cell surface by the positive charge of the cationic emulsions,increase the cell transfection effectively and is a convenient,economic and pragmatic gene administration method.
