响应曲面法优化超声波辅助提取苦荞总黄酮研究

【摘要】 利用响应曲面法对超声波提取苦荞总黄酮工艺条件进行优化。在单因素试验基础上,采用Plackett–Burman设计法筛选出影响总黄酮得率主要因素;再根据Box–Behnken中心组合设计原理对主要因素进行优化,建立超声波提取苦荞总黄酮二次多项式回归模型。结果表明:模型合理可靠,修正后超声波提取苦荞总黄酮最优条件为:乙醇浓度75%、提取温度73℃、液固比40∶1 ml/g、提取时间30 min、超声波功率160 W,在此条件下,总黄酮得率可达2.84%,与预测值2.86%基本符合。

【Abstract】 The optimal condition for ultrasonic–assisted extraction of total flavones of buckwheat was investigated by using response surface design.Based on the single factor experiments,firstly,Plackett– Burman design was applied to screen out the remarkable factors influencing the extraction of total flavones,then response surface methodology(RSM)was used to optimize the above critical factors,and the regression equation was also established.The results showed that the regression equation is reliable to analyse and predict the extracting process.The optimal extracting conditions are as follows:ethanol concentration 75%,ultrasonic temperrature 73℃,liquid–solid ratio 40 ml/g,time 30 min,power 160 W.Under such conditionst,he yield of total flavones is 2.84%i,n line with predictive value of 2.86%.
