金雀异黄素对胶质瘤细胞株U251MG凋亡的影响
Effects of Genistein on apoptosis of human glioma cell line U251MG
【摘要】 目的研究金雀异黄素(Genistein)对人胶质瘤U251MG细胞凋亡的影响及机制。方法碘化丙啶(propidium iodide,PI)和Annexin V-FITC双染色的流式细胞分析方法检测凋亡,并通过流式细胞仪和免疫细胞化学染色检测Bcl-2、bax蛋白的表达。结果 Genistein在25μg/ml和50μg/ml作用48 h后,早期凋亡率均较对照组明显增加(P<0.01),并呈剂量依赖性。Bcl-2蛋白表达较对照组明显下降(P<0.01),而bax蛋白则较对照组明显增加(P<0.01),呈剂量依赖性。结论 Genistein可诱导胶质瘤U251MG细胞凋亡,其机制可能与其上调bax蛋白以及下调Bcl-2蛋白的表达有关。
【Abstract】 Purpose To explore the effects of Genistein on the apoptosis of human glioma cell line U251MG and possible involved mechanism.Methods Cell apoptosis was examined by PI and Annexin V-FITC double staining with flow cytometry.The expression of Bcl-2 and bax proteins was examined with flow cytometry and immunocytochemistry.Results After treated with Genistein 25 μg/ml and 50 μg/ml for 48 h,the percentage of the apoptotic cells increased in a dose-dependent manner(P<0.01).Bcl-2 protein expression was significantly decreased and bax protein expression significantly increased in a dose-dependent manner as compared with control group(P<0.01).Conclusions Genistein can induce apoptosis of U251MG cells,and the mechanism may be associated with the upregulation of bax protein expression,and the downregulation of Bcl-2 proteins expression.
