艾蒿生长点及试管苗培养的研究

【摘要】 为了保护野生资源,满足人们的需求,以带有生长点的艾蒿茎段为材料,采用植物组织培养方法,对艾蒿生长点的分化培养、分化芽生根培养、试管苗移栽及定植进行了研究.研究结果表明:MS+NAA 1.0mg.L-1+GA3 0.5 mg.L-1+6-BA 3.0 mg.L-1+蔗糖30 g.L-1是艾蒿生长点分化培养的理想培养基;诱导分化芽生根的理想培养基是N6+IAA 0.2 mg.L-1+蔗糖20 g.L-1,且适宜在恒温光照条件下培养;试管苗移栽成活率高达100%,定植的试管苗保持了野生植株的生物学性状.

【Abstract】 The study aimed to protect wild resources and meet the need of people.In the experiment,the stems with growing point of Artemisia argyi were used as materials.By using tissue culture technique,growing point differentiation,rooting and transplanting of tube seedlings were studied.The results demonstrated that MS+NAA1.0mg L-1+GA30.5mg L-1+6-BA3.0mg L-1+sucrose30g L-1 was ideal medium for differentiation of growing point;the optimum medium for rooting of tube seedlings was N6+ IAA0.2mg L-1+ sucrose20g L-1,and it was suitable for cultivation under homothermal illumination;the survival rate of transplanted tube seedlings could be up to 100%,colonization of plantlets maintained for all biological traits which wild plant had.
