壳寡糖对体内外肝癌细胞生长的抑制作用及其机制探讨

【摘要】 目的探讨壳寡糖对体内外肝癌细胞生长的抑制作用及其相关机制。方法体外实验采用噻唑蓝(MTT)法检测壳寡糖对肝癌SMMC-7721细胞生长的影响,流式细胞术检测肝癌细胞凋亡率,RT-PCR方法检测Bcl-2 mRNA的表达。体内实验观察壳寡糖对肝癌H22移植瘤生长的影响,免疫组化检测肿瘤组织Bcl-2蛋白的表达。结果体外实验表明,壳寡糖能抑制肝癌细胞增殖和诱导肝癌细胞凋亡,其作用随剂量增加而增强;壳寡糖4mg/ml的细胞增殖抑制率为23.82%,壳寡糖2mg/ml的凋亡率为15.28%,与对照组比较,差异均有统计学意义(P<0.01);壳寡糖能下调肝癌细胞中Bcl-2 mRNA的表达。体内实验显示,壳寡糖能抑制小鼠肝癌H22移植瘤的生长,高剂量组300mg/(kg.d)的抑瘤率为33.13%,并可明显下调肿瘤组织中Bcl-2蛋白的表达。结论壳寡糖在体内外均具有抑制肝癌细胞增殖及诱导肝癌细胞凋亡的作用,其促凋亡的机制可能与下调Bcl-2的表达有关。

【Abstract】 Objective To investigate the inhibitory function of oligochitosan on hepatocellular carcinoma cells in vivo and in vitro.Methods In experiment in vitro,MTT assay was applied for evaluation of proliferation of SMMC-7721 cells treated by oligochitosan.Apoptotic cells were detected by flow cytometry and RT-PCR was used for the expression of Bcl-2.In experiment in vivo,we observed the effect of oligochitosan on the growth of H22 cell subcutaneous transplanting tumor in mice.Immunocytochemistry was utilized to measure the expression of Bcl-2 in the tumor tissues.Results Experiment in vitro showed oligochitosan inhibited SMMC-7721 cell proliferation and induced the apoptosis of them.This effect increased with the increasing dosage.The tumor inhibiting rate was 23.82% after treated with 4mg/ml of oligochitosan and the apoptosis rate was 15.28% with 2mg/ml of oligochitosan.Compared with those of the control group,the inhibitory effect was significant(P<0.01).The expression of Bcl-2 mRNA in SMMC-7721 cells was markedly suppressed by oligochitosan.In experiment in vivo,oligochitosan inhibited the growth of H22 tumor cells,the tumor inhibiting rate of high doses of oligochitosan(300mg/kg) was 33.13% and the expression of Bcl-2 was markedly suppressed in the tumor tissues by oligochitosan.Conclusion Oligochitosan both in vivo and in vitro inhibits the proliferation and induces the apoptosis of hepatocellular carcinoma cells.The mechanism is possibly that oligochitosan down-regulates the expression of Bcl-2.
