【摘要】 目的:探讨胆宁片对胆石症患者胆汁中33.5kDa泡蛋白含量及其结构的影响。方法:2008年1月—2010年3月对60例胆囊切除病例和40例胆总管探查病例进行前瞻性临床研究,其中60例胆囊切除病例随机分为3组:胆固醇结石胆宁片组(n=20)、胆固醇结石对照组(n=20)、胆色素性结石组(n=20),另外设立正常对照组(n=20)。通过酶联免疫吸附法检测每份胆汁中33.5kDa泡蛋白含量,比较各组之间的差异。40例胆总管探查病例分为研究组和对照组(n=20),通过蔓陀螺凝集素探针(DSA)介导的蛋白印迹法检测胆汁中33.5kDa泡蛋白糖链条带DSA结合率,比较2组糖链结构的变化。结果:胆固醇结石组胆汁中33.5kDa泡蛋白含量显著高于胆色素结石组和正常对照组,胆固醇结石胆宁片治疗组胆汁33.5kDa泡蛋白含量较对照组显著降低(P<0.01)。胆固醇结石治疗组泡蛋白糖链条带DSA结合率较对照组显著下降(P<0.05)。结论:胆宁片能降低胆固醇结石患者胆汁中33.5kDa泡蛋白含量,并通过改变糖链结构而使其促成核活性发生变化。更多还原

【Abstract】 Objective:To appraise the effect of Danning tablet on the concentration and structure of 33.5kDa vesicular protein in bile.Methods:From January 2008 to March 2010,60 cases of cholecystectomy were enrolled and randomly divided as Danning tablet treatment group with cholesterol gallstones: the control group with cholesterol gallstones(n=20),pigment gallstones group(n=20),a normal group(n=20).The concentration of vesicular protein in bile was examined by enzyme-linked immunosorbent assay(ELISA).In addition 40 cases of choledochotomy were divided into Danning tablet treatment group(n=20) and control group(n=20).The number of biliary 33.5 kDa vesicular protein sugar chain antennas of two groups were compared by Western blot analysis.Results:The biliary 33.5kDa vesicular protein in control group with cholesterol gallstones was significantly higher than those in other groups(P<0.01).The number of biliary 33.5 kDa vesicular protein sugar chain antennas in treatment group decreased significantly,and the binding rate of protein sugar chain declined too.Conclusions: Danning tablet can reduce the concentration of 33.5kDa vesicular protein in bile,change the structure of sugar chain and cause the pro-nucleating activity of the vesicular protein.更多还原
