【摘要】 目的观察内皮素受体拮抗剂C2010对内皮细胞系ECV304的作用,为进一步探讨其抗肿瘤作用提供依据。方法磺酰罗丹明B(SRB)方法评估内皮素受体拮抗剂C2010对内皮细胞系ECV304增殖的抑制作用;用流式细胞仪检测其对细胞凋亡的影响。结果 C2010在204、0μg/mL给药48 h后对ECV304细胞增殖有剂量依赖的显著抑制作用,抑制率分别为12.67%、43.32%,并能使ECV304细胞凋亡数量增加。结论 C2010对内皮细胞ECV304有抑制增殖作用和诱导凋亡作用。更多还原

【Abstract】 Objective To investigate the effects of the endothelin receptor antagonist C2010 on proliferation and apoptosis of ECV304 cell line and to provide some basis for further research on its anticancer effects.Methods Sulforhodamine B(SRB) assay was used to assess the effect of C2010 on proliferation of ECV304 cell line,and the cell apoptosis was detected by flow cytometry.Results The data showed that C2010(20,40μg/ml)could significantly inhibit the proliferation of ECV304 cells in a dose-dependent way after 48h,and the inhibition rate was 12.67%(P<0.05)and 43.32%(P<0.01),respectively.In addition,the apoptosis rate of ECV304 cells also increased significantly.Conclusion C2010 can effectively inhibit the proliferation and induce the apoptosis of ECV304 cells line in a dose dependent way.更多还原
