神经干细胞移植量对PST大鼠神经病理性疼痛的影响
Effect of different quantities of neural stem cells on partial sciatic nerve transaction induced neuropathic pain in rats
【摘要】 目的观察不同数量神经干细胞(NSCs)鞘内注射对坐骨神经半切断(PST)大鼠神经病理性疼痛及脊髓背角、背根神经节(DRG)胶质源性神经营养因子(GDNF)表达的影响。方法 84只成年雄性SD大鼠随机均分为七组:假手术组(Ⅰ组)、PST组(Ⅱ组)、PST+NSCs1×103组(Ⅲ组)、PST+NSCs1×104组(Ⅳ组)、PST+NSCs1×105组(Ⅴ组)、PST+NSCs1×106组(Ⅵ组)、PST+NSCs1×107组(Ⅶ组)。NSCs于术后3d鞘内注射。记录术前1d、术后1、3、7、14、21d机械痛及热痛阈;术后7、21d免疫组织化学与RT-PCR检测同侧脊髓背角及DRGGDNF表达。结果与Ⅰ组比较,Ⅱ~Ⅶ组术后1d痛阈下降,术后7、14d最低(P<0.05);与Ⅱ组比较,术后7dⅣ、Ⅴ、Ⅵ、Ⅶ组随着NSCs量的增加,痛阈逐渐升高,脊髓背角及DRGGDNF表达逐渐上调(P<0.05);术后21d,Ⅴ、Ⅵ、Ⅶ组痛阈均与术前差异无统计学意义,但其GDNF表达Ⅴ组最高(P<0.05)。结论鞘内注射1×105NSCs能最有效缓解PST大鼠神经病理性疼痛。
【Abstract】 Objective To observe the effect of different quantities of neural stem cells(NSCs)administered intrathecally on the behavior of neuropathic pain and expression of glial cell line-derived neurotrophic factor(GDNF)in the spinal cord dorsal horn and dorsal root ganglion in rats after partial sciatic nerve transaction(PST).Methods Eighty-four adult SD rats were randomly divided into seven groups:group sham(group Ⅰ),group PST(group Ⅱ),group PST+NSCs 1×103(group Ⅲ),group PST+NSCs 1×104(group Ⅳ),group PST+NSCs 1×105(group Ⅴ),group PST+NSCs 1×106(group Ⅵ),group PST+NSCs 1×107(group Ⅶ).NSCs were injected intrathecally on the 3rd postoperative day.Mechanical and thermal withdrawl thresholds were determined 1 d before operation and 1,3,7,14 and 21 d after operation.The expression of GDNF in the ipsilateral spinal cord dorsal horn and DRG were detected by immunohistochemistry and RT-PCR at the days 7 and 21 after PST.Results PST induced decrease in pain threshold since day 1 after nerve injury,and it reached lowest level after day 7(P<0.05).With the increase of intrathecal NSCs,mechanical and thermal withdrawl thresholds increased gradually and the expression of GDNF in both spinal cord dorsal horn and DRG increased correspondingly(P<0.05);On day 21,the mechanical and thermal thresholds in the groups Ⅴ,Ⅵ and Ⅶ had no significant changes compared with the group I,but the expression of GDNF in the group V was the highest among the groups(P<0.05).Conclusion Intrathecal NSCs 1 105 are the most effective in alleviating PST-induced neuropathic pain in rats.
