蒙药娜仁满都拉—11对Walker—256腹水型大鼠GATA—3和T—bet基因表达的影响
Effects of Na Ren Man Du La—11 on T—bet,GATA—3 Gene Expression in Walker—256 Ascite Rat
【摘要】 建立Walker—256腹水型肿瘤大鼠模型,娜仁满都拉—11灌胃。10 d后采集外周血(Peripheral Blood Mono-nuclearCell,PBMC),分离单个核细胞进行细胞培养,用RT-PCR法检测GATA—3、T—bet基因的表达水平。结果蒙药灌胃组比生理盐水对照组大鼠生存质量明显改善,GATA—3表达降低,T—bet表达增高(P<0.05),提示蒙药娜仁满都拉—11能通过增强Th1细胞特异性转录因子T—bet的表达,降低Th2细胞特异性转录因子GATA—3的表达,发挥抗肿瘤免疫调节作用。
【Abstract】 Duplicate walker—256 ascite model in rat,study the anti-tumur effect of Na Ren Man Du La—11 and influence on T—bet,GATA—3 gene expression.The rat bearing tumor were divided into treatment group and control group,535 mg/(kg·d) Na Ren Man Du La—11 were given to the mice in treatment group and 0.9%NaCl were given to those in negative control group.10 days later,the peripheral blood were collected for separated mononuclear cell and these mononuclear cell were cultivated.RT—PCR were used to test quantity the expression of T—bet and GATA—3 gene on these cells.The Results suggested that Na Ren Man Du La—11 could obviously improve the life qualities of mice.The expression of T-bet was lower in treatment group than in negative control group,while The expression of GATA—3 was higher than in control group(P<0.05).Thus,Na Ren Man Du La—11 has anti-tumor function in treating ascite model.It also has function in immune adjustment by enhance the expression of GATA—3 and lower the expression of T—bet in ascite model.
