HPLC法测定肿瘤患者外周血5-FU血药浓度

【摘要】 目的建立肿瘤患者外周血5-FU血药浓度的高效液相色谱法(HPLC)测定方法.方法采用HPLC法测定肿瘤患者血浆中5-FU的血药浓度.色谱柱:Xterra RPC18(150 mm×4.6 mm,5μm);流动相:纯水;流速为1.0 mL/min,检测波长为265.2 nm.结果 5-FU线性范围为(0.125~9.0)μg/mL,线性方程为:Y=1.48×10-2X+7.67×10-3,r=0.999 7(n=7),批内RSD小于9.8%,批间RSD小于9.6%,方法学回收率为99.34%~107.86%.结论本方法可用于肿瘤患者外周血5-FU血药浓度的常规监测.

【Abstract】 Objective To establish an HPLC method for determining the concentration of 5-Fluorouraci in blood of cancer patients.Methods The concentration of 5-FU in blood of patients with cancer was determined by HPLC.The liquid chromatography column was Xterra RPC18(150 mm×4.6 mm,5 μm).the mobile phase was water.The flow rate was 1.0 mL/min.UV detection wavelength was at 265.2 nm.Results A linearity was obtained from(0.125 to 9.0) μg/mL for 5-FU.The calibration equation of 5-FU was Y=1.48×10-2X+7.67×10-3,r=0.999 7(n=7).The RSD within day and between days were less than 9.8% and 9.6% for 5-FU,respectively.The recovery rate of 5-FU was 99.34%~107.86%.Conclusion This method can be used for routine clinical determination of the concentration of 5-FU in blood of cancer patients.
