贵州野生与栽培天麻种质资源的SSR分析

【摘要】 目的对贵州省野生和栽培共24个天麻基因组DNA的遗传多态性进行分析,构建其SSR指纹图谱。方法采用改良的CTAB法提取天麻基因组DNA,采用NTSYSpc软件进行UPGMA聚类分析和遗传相似度分析。结果改良的CTAB法能够获得纯度和浓度较高的天麻基因组DNA,PCR扩增获得较清晰的SSR指纹图谱,UPGMA聚类可明显区分野生与栽培天麻。结论 SSR指纹图谱的UPGMA聚类分析表明野生天麻和栽培天麻遗传相似度较接近,栽培天麻产生较大变异。

【Abstract】 Objective Get genomic DNA from Genetic Gastrodia elata Blume.for PCR,analyzed genetic polymorphisms of genomic DNA from 24 samples of wild and cultivated Gastrodia elata Blume.in Guizhou,set up the SSR fingerprintings.Methods DNA was obtained by CTAB methods,and 10 primers from Genebank were selected for SSR-PCR,UPGMA dendrogram was analyzed by NTSYSpc with SSR fingerprintings.Results Genetic DNA obtained by CTAB mehods with high purity and cencetration was fit for PCR,got 3 primers from 10 and clear fingerprintings,the similarity coefficient of the fingerprintings can obviously distinguish the wild and cultivated Gastrodia elata Blume..Conclusion UPGMA dendrogram of SSR showed that wild and cultivated Gastrodia elata Blume.could be distinguished from each other,but cultivated one showed the great genetic viaration.
