三带模拟移动床对芍药苷及芍药内酯苷异构体的分离
Isomers Separation Methods of Paeoniflorin and Albiflorin by Three-zones SMB
【摘要】 介绍了应用三带模拟移动床分离得到高质量分数芍药苷和芍药内酯苷异构体的工艺。确定了模拟移动床分离芍药苷和芍药内酯苷的操作条件,SMBC运行模式:1-1-2,样品质量浓度:40 g/L,进样流速:UF=0.3 mL/min,洗脱流速:UD=2 mL/min,萃取流速:UP=4 mL/min,切换时间:ts=25 min,洗脱液及萃取液:V(甲醇)∶V(水)=30∶70。产品中芍药苷的质量分数根据标准曲线计算为92%,收率为85%;芍药内酯苷的质量分数为94%,收率为87%。讨论了影响模拟移动床分离的主要因素。
【Abstract】 The isomer separation process of high mass fraction paeoniflorin and albiflorin by the three-zones SMB was described.The working parameters of SMB were set as follows:mode:1-1-2;injection mass concentration:40 g/L;flow rate of injection:0.3 mL/min;flow rate of elution:2 mL/min;flow rate of extract:4 mL/min;switch time:25 min;mobile phase:V(methanol)∶V(water)=30∶70.The paeoniflorin mass fraction in extraction is 92% by the standard curve method,and the recovery is 85%;the albiflorin mass fraction in raffinate is 94%,and the recovery is 87%.Simultaneously,the main factors which affect the separation process in SMBC were discussed.
