葛根素对大鼠肝缺血再灌注后核因子-κB和肿瘤坏死因子-α表达的影响

【摘要】 目的:探讨葛根素对大鼠肝脏缺血再灌注损伤中NF-κB及TNF-α表达的影响。方法:对大鼠肝脏建立部分缺血再灌注模型,以葛根素预处理,应用免疫组化方法测定肝组织中NF-κB的活化程度及TNF-α的表达,测定肝脏丙二醛(MDA)含量并对肝组织行常规病理学检查以及电镜观察。结果:大鼠肝脏缺血再灌注损伤后肝组织NF-κB活化程度、TNF-α的表达以及MDA表达升高,经葛根素预处理后,肝组织NF-κB、TNF-α活化表达程度及MDA显著下降,且病理改变较轻。结论:葛根素能够减少氧自由基产生,抑制NF-κB的活化及表达,减少TNF-α的产生,对肝脏缺血再灌注损伤有保护作用。

【Abstract】 Objiective:To investigate the expressions of nuclear factor-kappa B(NF-κB) and Tumor Necrosis Factor-a(TNF-α) in hepatic ischemia-reperfusion injury of SD rats and the protective effect of Puerarin.Methods: A rat model of partial hepatic ischemia-reperfusion was made.By using Puerarin pretreatment,the NF-κB and TNF-a expression were measured by immunohistochemical method and the levels of Maleic Dialdehyde(MDA) were examined.The morphology of liver tissue was observed under pathological examination and electron microscope.Results: After Reperfusion,the expression of NF-κB and TNF-α increased significantly.MDA levels,NF-κB and TNF-α expression of the I/R group were significantly increased after reperfusion.MDA,NF-κB and TNF-α expression of the pue group were significantly lower than those of the I/R group(P<0.05),and hepatic morphological injury was slighter.When treated with puerarin,the expression of NF-κB and TNF-α decreased obviously and the pathological changes were also lessened.Conclusions: Puerarin has preventive effects against hepatic ischemia-reperfusion injury by reducing the production of the free radical,inhibiting the activiation and expression of NF-κB,reducing the production of TNF-α.Puerarin can protect the liver from the ischemia reperfusion injury.
