肠道病毒71型(EV71)ICR乳鼠动物模型的建立

【摘要】 目的建立ICR乳鼠动物模型,为抗EV71药物的筛选和活性评价提供一定的信息支撑。方法采用EV71国际标准株BrCr株,分别对1日龄、7日龄及21日龄ICR乳鼠经腹腔注射感染,并定期采集感染小鼠器官组织进行病原学诊断,确定EV71的感染情况。结果经腹腔途径感染1日龄和7日龄ICR小鼠出现精神萎靡、肢体麻痹瘫痪、消瘦、死亡等症状。腹腔注射分别在1日龄乳鼠肺、脑、肠及7日龄乳鼠肺、脑、肠、大腿肌肉检测到病毒RNA,而21日龄乳鼠出现轻微症状后自行恢复。本研究同时建立了一种分子生物学方法,为今后研究其他适合EV71的动物模型奠定了基础。结论成功建立EV71感染动物模型。发现不同日龄ICR乳鼠对EV71国际标准株感染的易感能力和感染后发病程度不同,低龄(1日龄和7日龄)乳鼠易感,而高龄(21日龄)乳鼠不易感染。ICR乳鼠可用于EV71病毒感染机制、病毒体内分布等基础研究,并可作为抗EV71病毒药物筛选和活性评价动物模型应用,但模型的稳定程度尚需继续研究。

【Abstract】 Objective To create possible enterovirous 71(EV71)ICR mice animal models for the identification of the pathogenetic mechanism and vaccine evaluation.Method ICR mice of different age were infected with EV71 virus through the intraperitoneal(IP)route.The organ and tissue of the dying mice were collected at various time points for pathogeny diagnoses.Meanwhile,one step RT-PCR method was developed in our study.Results The ICR mice infected through the intraperitoneal(IP)route showed severe clinical symptoms,such as cachexia,low movement,paralysis,low body mass and death.Virus RNAs could be detected by RT-PCR in the lung,brain and intestine in the neonate mice that were infected through the IP route.Virus RNAs were detected by RT-PCR in the lung,brain,intestine and leg muscle in the seven-day mice.The moleculer biology method developed could be used for the further study of other animals in the future.Conclusion The infection of ICR mice(one-day mice,one-week mice,or three-week mice) through the IP route proves to be at different levels using EV71 strain isolated from patients.The outcome of infection of ICR mice can be used for pathogenetic research as an animal model despite some instability.
