乌桕优选单株的离体快速繁殖技术研究
In vitro culture and plant regeneration of Sapium sebiferum
【摘要】 选择8年生乌桕优树母株,取其枝条的腋芽茎段为外植体,进行组织培养快速繁殖技术研究。结果表明,接种时,先用70%的酒精对带芽茎段消毒30 s,再用0.1%升汞灭菌11 min,污染率可降到18.5%。在初代培养中,芽诱导最佳培养基为WPM+6-BA 1.5 mg/L+NAA 0.2 mg/L,30 d后的诱导率可达85.4%。在4/5 MS+6-BA1.5 mg/L+KT 0.5 mg/L+NAA 0.05 mg/L培养基中继代增殖培养,增殖效果最好,增殖倍数为3.8,生长高度达4.8 cm。将健壮的试管芽苗接种于1/2 MS+IBA 0.5 mg/L+NAA 0.3 mg/L+Citric acid 100 mg/L培养基上进行生根诱导,生根率可达95.8%。移栽35 d后,成活率达87.8%。
【Abstract】 The stem sections with axillary bud of Sapium sebiferum(L.) Roxb.(8 years old) were used as explants for the research of tissue culture and rapid propagation.The results showed that the contamination rate was 18.5% when we sterilized the explants with ethanol(70%) 30 seconds then used HgC12(0.1%) to sterilize the stem sections 11 minutes.The medium suitable for induction in primary culture was WPM+6-BA 1.5 mg/L+NAA 0.2 mg/L,and the inductivity reached to 85.4% after 30 days.The medium suitable for secondary proliferation was 4/5 MS+6-BA 1.5 mg/L+KT 0.5 mg/L+NAA 0.05 mg/L.The proliferation coefficient was 3.8 and the bud length could reach to 4.8 cm.When the sturdy test-tube plantlets were inoculated into the medium of 1/2 MS+IBA 0.5 mg/L+NAA 0.3 mg/L+Citric acid 100 mg/L for rooting induction,the rooting rate became over 95.8%.The transplanting survival rate was 87.8% after 35 days.
