人参皂甙Rb1促进老年大鼠睾丸间质细胞睾酮产生和StAR蛋白表达
Ginsenoside Rb1 enhanced testosterone secretion and expression of StAR in aged rat leydig cells
【摘要】 目的:探讨人参皂甙Rb1对老年大鼠间质细胞增殖及睾酮产生的影响,分析人参皂甙Rb1对睾酮提升作用的机制。方法:体外分离培养老年大鼠睾丸间质细胞,培养液中分别加入不同浓度Rb1,MTT法测定细胞增殖改变,ELISA法测定培养上清中睾酮水平,用RT-PCR分析间质细胞StAR基因和P450scc基因表达,western blotting方法检测间质细胞StAR和P450scc蛋白表达水平。结果:人参皂甙对间质细胞增殖没有明显影响,老年大鼠睾丸间质细胞分泌睾酮水平明显低于青年大鼠,人参皂甙Rb1呈浓度依赖性上调老年大鼠睾丸间质细胞睾酮产生,上调StAR基因和蛋白表达水平,但对P450scc基因和蛋白表达基本没有影响。结论:衰老致睾丸间质细胞睾酮产生能力下降,人参皂甙单体Rb1可通过上调StAR蛋白表达,增强睾酮合成,达到延缓衰老的目的。
【Abstract】 Objective:To observe the effect of ginsenoside Rb1 on testosterone production of aged rat leydig cells and to explore the mechanism of Rb 1 restores the ability of aged leydig cells to produce testosterone at the high levels characteristic of young cells.Methods:Leydig cells isolated from young and old rats were cultured for 2d with or without Rbl,then the culture supernatant were collected for quantitation of testosterone by ELISA and leydig cells were harvested for detection of steroidogenic acute regulatory protein and cytochrome P450 side chain cleavage mRNA and protein expression levels by RT-PCR and Western blotting,respectively.Results:ELISA analysis revealed that the testosterone levels in culture supernatant of aged leydig cells were higher than that of young leydig cells,while Ginsenoside Rbl could stimulate testosterone ,production and steroidogenic acute regulatory protein expression in aged leydig cells in a dose-dependent manner.Conclusion:With aging,the capacity of the leydig cells to produce testosterone declines significantly.Ginsenoside Rbl could enhance its steroidogenic function by up-regulating the expression of steroidogenic acute regulatory protein.
