牛乳铁蛋白卵黄抗体的制备及纯化方法的建立

【摘要】 通过自制并初步纯化牛乳铁蛋白的卵黄抗体,用肌肉和皮下注射结合的方法罗曼蛋鸡进行免疫来制备多克隆抗体。选用150日龄的罗曼蛋鸡4只进行不同浓度的多次皮下免疫注射,采集卵黄抗体并用硫酸铵沉淀法进行抗体的初步纯化,经四次免疫后的罗曼蛋鸡卵黄抗体的效价分别达到了l∶32 000和l∶24 000,并在较高水平的情况下维持两周以上,同时经初步纯化后抗体纯度得到了很大提高。试验结果显示肌肉和皮下注射结合的方法对罗曼蛋鸡进行免疫来制备多克隆抗体是可行的,同时通过硫酸铵沉淀法纯化抗体可以达到试验的需求。

【Abstract】 Through self-preparing and purifying the egg yolk antibody of bovine lactoferrin,the polyclonal antibody was prepared by combining the methods of muscle and subcutaneous to immune the Roman laying hens.Four Roman laying hens at 150d ages were selected to conduct the subcutaneous immunization several times with different densities.The egg yolk antibodies were collected and purified preliminarily with the method of ammonium sulfate precipitation.After four times of immunization,the egg yolk antibody titer of Roman laying hens arrived at 1∶32 000 and 1∶24 000 and lasted for over two weeks at high levels,and the purity of the antibody after preliminary purification improved greatly.The results show that it is feasible to prepare polyclonal antibody through the method of combination of muscle and subcutaneous immunization and it can meet the needs of experiment with the method of ammonium sulfate precipitation.
