颞叶内侧型癫痫大鼠模型海马GABAA受体亚单位α1 β3 γ2表达研究

【摘要】 目的通过测定颞叶内侧型癫痫大鼠模型海马的GABAA受体的三个亚单位α1β3γ2的mRNA表达水平,来探讨它们在该病的发病机制和发展过程中的作用,为癫痫的病因学研究和临床治疗提供可能的理论依据。方法用锂和匹罗卡品腹腔注射20只成年大鼠,制作颞叶内侧型癫痫大鼠模型。用断头法完整采集大鼠海马,RT-PCR技术扩增三个亚单位α1β3γ2的mRNA,通过t-检验与正常大鼠进行对比。结果实验组有16只大鼠制作癫痫模型成功,与β-actin电泳条带比值:α1(0.369±0.164),β3(0.499±0.273)γ2(0.429±0.346);对照组分别为α1(0.518±0.212),β3(0.291±0.116),γ2(0.231±0.132)。t-检验结果P<0.05,存在统计学差异。结论实验组大鼠GABAA受体可能发生了重组,它的三个亚单位α1β3γ2可能参与了颞叶内侧型癫痫的发病机制。

【Abstract】 Objective The purpose of this research is approaching the contribution of GABAA receptor in the genisis and development of MTLE through determing the mRNA level of α1、β3 and γ2 subunit.So as to it may has some breakthrough for etiology and clinical therapy of epilepsy.Methods Totally 20 MTLE rat models are injected with lithium and pilocarpine intraperitoneally.They are all taken out of hippocampus totally operation.The cDNA is obtained through reverse transcription and the mRNA of α1、β3 and γ2 subunits through polymerase chain reaction.The ratio of experimental and control group are analysed by t-test through SPSS software to determine if they are different.Results Totally 16 rats of experimental group maintain epileptic status successfully.The ratios of experimental group are: α1(0.369±0.164),β3(0.499±0.273)γ2(0.429±0.346);The ratios of control group are: α1(0.518±0.212),β3(0.291±0.116),γ2(0.231±0.132).The P values are all less than 0.05.The are significantly different instatistical analysis.Conclusions The GABAA receptors have recombinated,α1、β3 and γ2 subunits may play a role in the etiology of MTLE.
