天智颗粒对血管性痴呆大鼠海马Tau蛋白、β淀粉样蛋白及VEGF表达的影响

【摘要】 目的研究天智颗粒对血管性痴呆(vascular dementia,VD)大鼠海马Tau蛋白、β淀粉样蛋白(amyloidβ-protein,Aβ)及血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响。方法将135只大鼠随机分为3组,模型组,假手术组和治疗组。采用双侧颈总动脉结扎法制备VD大鼠模型,治疗组应用天智颗粒5g/kg灌胃,模型组和假手术组应用等量蒸馏水灌胃,1次/d,在相应时间点通过免疫组织化学法测定各组大鼠海马CA1区Tau蛋白、Aβ及VEGF表达。结果治疗组大鼠1个月和2个月时Tau蛋白表达与模型组相比明显下调,差异有统计学意义(P<0.05);3组Aβ表达两两比较差异无统计学意义(P>0.05);治疗组各个时间点VEGF表达较其余两组明显上调,差异有统计学意义(P<0.05),治疗组2个月时表达最高,各亚组之间比较差异有统计学意义(P<0.05)。结论天智颗粒可抑制Tau蛋白磷酸化,使VEGF表达上调,对Aβ的表达无明显影响。

【Abstract】 Objective To investigate the effects of Tianzhi particle on Tau protein,amyloid β-protein(Aβ)and vascular endothelial growth factor(VEGF)on hippocampus tissue of vascular dementia model rats.Methods Totally 135 rats were randomly divided into model group,sham operation and treatment group.Model of vascular dementia was established by bilateral common carotid artery occlusion.In the treatment group,we use Tianzhi particle to perfuse the stomach(5g/kg).In the model group and sham operative group,we use distilled water(5g/kg)to perfuse the stomach.The expression of phosphorylated Tau protein,Aβ and VEGF in hippocampal CA1were detected by immunohistochemistry at 7 days,1months and 2months.Results Compared with model group,the expression of Tau protein decreased apparently at 1month and 2 month(P<0.05),there were no significant difference in the expression of Aβ among three groups(P>0.05).Compared with model group and sham operation group,the expression of VEGF was highly improved(P<0.05).Conclusions Tianzhi particle can not only inhibit the phosphorylation of Tau protein,but also upregulate the expression of VEGF.However,Tianzhi particle has no effects on the expression of Aβ.
