IFN-γ对白血病U937细胞株B7-H1表达的影响
Effect of IFN-γ on B7-H1 expression of leukemia cell line U937
【摘要】 目的探讨IFN-γ对白血病U937细胞株B7分子同源配体1(B7 homolog 1,B7-H1)表达的影响。方法采用不同剂量的IFN-γ处理U937细胞株48h后,分别采用免疫组化法、流式细胞术和实时荧光定量RT-PCR检测IFN-γ对U937细胞表达B7-H1的影响。结果①与对照组(IFN-γ0 ng/ml)比较,在蛋白水平,经不同浓度的IFN-γ(50ng/ml、100ng/ml和150ng/ml)处理后的U937细胞B7-H1表达强度明显升高(P<0.001);而且B7-H1的表达水平与IFN-γ之间存在剂量效应关系。②在mRNA表达水平,IFN-γ能以剂量依赖性方式诱导U937细胞的B7-H1表达:IFN-γ50ng/ml、100ng/ml和150ng/ml组B7-H1的表达水平较对照组分别升高了6.5、26和22.7倍。结论 IFN-γ以剂量依赖性方式上调U937细胞上B7-H1表达。
【Abstract】 Objective To investigate the effect of interferon-γ(IFN-γ) on B7-H1 expression of leukemia cell line U937.Methods After being treated with different doses of IFN-γ for 48 hours,B7-H1 expression of U937 cell was detected by immunohistochemistry,flow cytometry and real-time quantitative RT-PCR.Results Compared with that of control group(IFN-γ,0ng/ml),the levels of protein expression of B7-H1 were significantly higher after being treated by IFN-γ(50ng/ml,100ng/ml and 150ng/ml)(P<0.001) in dose dependent manner.IFN-γ could up-regulate the mRNA expression of B7-H1 on U937 in dose dependent manner.The B7-H1 gene expression of IFN-γ(50ng/ml、100ng/ml and 150ng/ml) groups were 6.5 times,26 times and 22.7 times higher than that of control group respectively.Conclusions IFN-γ could remarkably up-regulated the B7-H1 expressions of leukemia cell line U937 in dose dependent manner.
