邻菲罗啉衍生物与G-四链体DNA的识别
Recognition of phenanthroline derivative for G-quadruplex DNA
【摘要】 设计合成了一种新型邻菲罗啉衍生物,用IR1,H-NMR1,3 C-NMR和质谱进行了结构表征。在包含100mM NaCl的Tris-HCl(pH 7.4)缓冲溶液中,利用荧光共振能量转移熔点(FRET-melting)分析以及紫外可见光谱研究了此化合物对人端粒G-四链体DNA(AG3(T2AG3)3)的稳定作用。发现2.0μM的化合物可以使G-四链体的熔点温度升高(ΔTm)7.9 oC,表明新合成的化合物具有识别并稳定人端粒G-四链体DNA的作用,二者的结合常数为(2.0±0.06)×106 M-1。
【Abstract】 A new phenanthroline derivative was synthesized and the product was characterized by means of IR,1H-NMR,13C-NMR and ESI-MS spectrometries.Stabilization effect of the target compound on the human telomeric G-quadruplex DNA(AG3(T2AG3)3) was further studied using FRET melting assay and UV-visible spectra in 10 mM Tris-HCl buffer(pH 7.4) containing 100 mM NaCl.The result showed an increase in the melting temperature(ΔTm) of 7.9 oC for G-quadruplex in the presence of 2.0 μM compound,suggesting that this compound can recognize and stabilize G-quadruplex structure,and their association constant is(2.0±0.06)×106 M-1.
