中药活性成分山萘酚与牛血清白蛋白相互作用的研究
Studies on the Interaction between Kaempferol and Bovine Serum Albumin
【摘要】 应用光谱法研究了中药活性成分山萘酚(Kaempferol)与牛血清白蛋白(BSA)的相互作用,探讨了山萘酚对BSA的荧光猝灭过程的机理,测定了不同温度下该结合反应的结合模式、结合常数、结合位点数、结合热力学参数等;并利用经典位点探针试剂华法林(Warfarin)和布洛芬(Ibuprofen)研究了山萘酚与BSA作用的具体部位在位点Ⅱ。
【Abstract】 In this study,the binding of Kaempferol to bovine serum albumin(BSA) was studied at physiological conditions(pH=7.40) by fluorescence and UV-Vis spectroscopies.Kaempferol strongly quenches the intrinsic fluorescence of BSA,and a decrease in the fluorescence quenching constant is observed together with an increase in temperature,which indicates that the fluorescence quenching of BSA by Kaempferol is a result of the formation of Kaempferol-BSA complex.The binding parameters,binding mode,and binding site were obtained.Furthermore,site marker competitive displacement experiments demonstrating that Kaempferol binds with high affinity to site II(subdomain IIIA) of BSA.
