聚氨酯不同低温等离子体改性及生物相容性评价

【摘要】 目的为构建组织工程化血管提供组织相容性好的支架材料。方法聚氨酯膜分别用CO2,O2,NH3和SO24种低温等离子体辐照进行改性,每种等离子体分别辐照2,5,10,15 min,采用生物学与材料学方法对改性前、后聚氨酯表面的理化特性及组织相容性进行比较。结果 CO2,O2及NH3等离子体改性后与未改性聚氨酸膜外观相同,倒置相差显微镜观察、电子显微镜扫描及甲苯胺蓝染色显示细胞在CO2,O2及NH3等离子体改性聚氨酸膜上生长状况较改性前好,O2辐照10 min的聚氨酸膜上细胞生长状况较其他等离子体及其他辐照时间好(P<0.05);改性后材料的表面粗糙度有所增加,并成功引入-OH-、COOH-、NH2-、SO3H等极性基团,这些基团明显改善聚氨酯表面的组织相容性;与改性前及其他方法比较,O2等离子体辐照10 min的聚氨酯组织相容性明显改善。结论低温等离子体是一种有效改善聚氨酯组织相容性的方法;O2等离子体辐照10 min的改性效果良好。

【Abstract】 Objective To provide scaffold material with good tissue compatibility for constructing tissue engineering blood vessel.Methods Low-temperature plasma of CO2,O2,NH3 and SO2 were used for 2,5,10 and 15 minutes respectively to treat the surface of polyurethane.The changes of physical and chemical characteristics and tissue compatibility of the polyurethane’s surface were evaluated and compared by biologic and material methods.Results Polyurethane membranes treated with CO2,O2,and NH3 plasma had the same appearance with the untreated polyurethane.Inverted phase contrast microscope,scanning electron microscope and toluidine blue staining test showed cells grew better after CO2,O2,NH3 plasma irradiation than those before treatment,and cells on the membrane irradiated by O2 for 10 minutes grew better than those irradiated by other plasmas for other irradiation periods.MTT assay showed O2 irradiation for 10 minutes had a higher optical density value compared with the other plasma treated membranes and the untreated membrane(P<0.05).After treatment,the surface roughness increased and some polar groups such as-OH,-COOH,-NH2 and-SO3H were successfully introduced to the surface,which significantly improved tissue compatibility of polyurethane’s surface.The improved compatibility of polyurethane treated by O2 plasma for 10 minutes was the best.Conclusion Low-temperature plasma is an effective method to improve the tissue compatibility of polyurethane and O2 irradiation for 10 minutes is the optimum treating condition.
