槲皮素对高尿酸血症大鼠黄嘌呤氧化酶和腺苷脱氨酶活性的影响
Effects of quercetin on activity of XOD and ADA in hyperuricemic rats
【摘要】 目的:研究不同剂量槲皮素(Que)对腺嘌呤+盐酸乙胺丁醇诱导高尿酸血症大鼠的影响及其机制。方法:利用腺嘌呤+盐酸乙胺丁醇片灌胃法诱导SD大鼠制备高尿酸血症模型,Que预防治疗3周,测定血清尿酸(SUA)、血清以及肝脏中的黄嘌呤氧化酶(XOD)、腺苷脱氨酶(ADA)活性。结果:各剂量Que均可降低SUA水平(F组间=1351.524,F时间=420.064,F交互=431.211,P均<0.001),血清XOD和ADA活性降低(F分别为17.588和16.294,P均<0.001),肝脏XOD和ADA活性亦降低(F分别为5.922和13.183,P均<0.001)。结论:Que可能通过抑制肝脏中XOD和ADA的活性来减少尿酸的生成,从而降低血尿酸。
【Abstract】 Aim:To study the effects and mechanism of different doses quercetin(Que) on hyperuricemic rats which were lavaged by adenine and ethambutol hydrochloride.Methods:SD rats with hyperuricemia were induced by adenine and ethambutol hydrochloride intragastrically,and then given Que for 3 weeks.The serum uric acid(SUA) and the activity of serum and liver xanthine oxidase(XOD),adenosine deaminase(ADA) were observed.Results:Every dose of Que could effectively reduce SUA(Fgroup=1 351.524,Ftime=420.064,Fgroup×time=431.211,P<0.001).Serum XOD and ADA activity was significantly lower than those of control group(F=17.588 and 16.294,P<0.001),and the activity of XOD,ADA in the liver of hyperuricemic rats were also significantly lower(F=5.922 and 13.183,P<0.001).Conclusion:Que may inhibit the activity of XOD and ADA in the liver and then the formation of uric acid reduces and SUA is lower.
