枸杞多糖对荷H22肝腹水瘤小鼠免疫功能的影响
Effects of lycium barbarum polysaccharides on immune function of H22 tumor bearing mice
【摘要】 目的:观察枸杞多糖(LBP)对荷H22肝腹水瘤小鼠免疫功能的影响。方法:昆明小鼠40只,分成4组(n=10),腹腔注射200μL的H22肝腹水瘤细胞(106mL-1),24h后分别以5、10或20mg/(kg·d)的LBP和等体积生理盐水灌胃,记录各组小鼠成瘤期和生存期。另取40只,随机分成4组(n=10),同上处理,连续喂养14d后处死小鼠,分别计算胸腺指数和脾指数;分离小鼠骨髓树突状细胞(DC),流式细胞术检测CD86、CD11a的表达,Westernblot检测白介素12(IL-12)P40的表达;诱导培养小鼠脾脏T淋巴细胞,MTT法检测分别以4组小鼠骨髓DC致敏的T淋巴细胞对H22肿瘤细胞的杀伤活性。结果:LBP可延缓荷瘤小鼠的成瘤期,延长其生存期(χ2=28.808和35.410,P均<0.001);提高其胸腺指数和脾指数(F=262.396和126.976,P均<0.001);使骨髓DCCD86、CD11a和IL-12P40的表达升高(F=3122.694,1469.946和252.590,P均<0.001);DC致敏的T淋巴细胞杀伤活性增强(F=122.539,P<0.001)。结论:LBP可以促进荷H22肝腹水瘤小鼠骨髓DC的成熟。
【Abstract】 Aim:To investigate the effects of lycium barbarum polysaccharides(LBP) on the immune function of H22 tumor bearing mice.Methods:A total of 40 mice were allocated into 4 groups at random(n=10).Then 200 μL H22 cells(106 mL-1) were injected into the left abdomen of the mice and 24 h later the mice were fed with LBP at the doses of 5,10,and 20 mg/(kg·d) as well as the equal volume of normal saline(control group) respectively.The survival time and the time of the tumor formed were recorded.Another forty mice were allocated into 4 groups as above and after 14 days,the mice were put to death,and the thymus index and splenic index were calculated.The mice bone marrow-derived DCs were separated,and the expressions of CD86 and CD11a and that of IL-12P40 were detected using MTT and Western blot,respectively.The spleen-derived T lymphocytes were induced with DCs from the 4 groups and their capabilities of killing H22 cells was assayed using MTT method.Results:Compared with control,the tumor formation time and the survival time of H22 tumor bearing mice in LBP groups prolonged(χ2= 28.808 and 35.410,P<0.001);the thymus index and splenic index were elevated(F=262.396 and 126.976,P<0.001),the expressions of CD86,CD11a,and IL-12P40 were up-regulated too(F=3 122.694,1 469.946,and 252.590,P<0.001).At the same time,the killing capabilities of T lymphocytes increased(F=122.539,P<0.001).Conclusion:LBP has positive effects on the immune function of H22 tumor bearing mice.
