白杨素敏化TRAIL诱导人胃癌SGC-7901细胞凋亡
Sensitization of TRAIL-induced apoptosis in human gastric cancer SGC-7901 cell line by chrysin
【摘要】 目的:研究白杨素(ChR)是否具有增敏肿瘤坏死因子相关凋亡诱导配体(TRAIL)诱导人胃癌SGC-7901细胞凋亡作用。方法:体外培养SGC-7901细胞。碘化丙啶(PI)染色流式细胞术(FCM)分析细胞凋亡率,琼脂糖凝胶电泳检测DNA梯形条带。结果:ChR(40μmol/L)、TRAIL(100 ng/mL)以及两者合用的细胞凋亡率分别为(4.57±0.54)%、(3.70±0.17)%和(48.81±4.11)%。两者合用后,琼脂糖凝胶电泳呈现细胞凋亡的典型DNA梯形条带。结论:白杨素具有敏化TRAIL诱导人胃癌SGC-7901细胞凋亡作用。
【Abstract】 Objective To investigate whether chrysin(ChR) sensitizes TRAIL-induced apoptosis in human gastric cancer SGC-7901 cell line.Methods Human gastric cancer SGC-7901 cell line was cultured in vitro.The cell apoptosis rate was examined by flow cytometry using PI fluorescence staining.DNA fragmentation was detected by DNA agarose gel electrophoresis.Results The apoptosis rate in human gastric cancer SGC-7901 cells treated by ChR(40μmol/L) or TRAIL(100ng/mL) or both were(4.57±0.54)%,(3.70±0.17)% and(48.81±4.11)%,respectively.The obvious fragmentation DNA was observed in combination of ChR and TRAIL.Conclusion ChR at suboptimal concentration sensitizes TRAIL-induced apoptosis in human gastric cancer SGC-7901 cells.
