吸毒合并HIV感染与HLA-DRB1*1301/1302等位基因的相关性

【摘要】 目的探讨吸毒并HIV感染与HLA-DRB1*1301/1302等位基因的相关性,为寻找吸毒合并HIV感染的易感基因和抵抗基因提供线索。方法应用序列特异性引物聚合酶链反应(polymerase chain reaction with sequence-specific primers,PCR-SSP)技术对41例无亲缘关系的吸毒合并HIV感染者及24例单纯吸毒者进行HLA-DRB1*1301/1302等位基因进行检测,分析正常对照组和吸毒组及吸毒合并HIV感染组HLA-DRB1 1301/1302等位基因频率分布。结果正常对照组*1301/1302等位基因频率为0.06573。吸毒组等位基因频率为0.04167,RR=0.600649,与正常组比较有下降趋势,但无统计学意义(P>0.05);吸毒合并HIV感染组等位基因频率为0.01220,RR=0.1700,与正常组比较有显著下降趋势(P<0.05)。吸毒组与吸毒合并HIV感染组等位基因频率无统计学差异(P>0.05)。结论 HLA-DRB1*1301/1302等位基因动态性与吸毒合并HIV感染的抵抗性密切相关,这对阐明疾病的病理、治疗和免疫预防具有重要作用。

【Abstract】 Objective To investigate the relationship between HLA-DRB1*1301/1302 allele polymorphism and HIV infection in drug users,and provide a cue for searching the susceptible allele and resistant allele.Methods A total of 41 drug users with HIV infection and 24 drug users without genetic relationship were studied.HLA-DRB1*1301/1302 allele polymorphism were tested by polymerase chain reaction with sequence-specific primers(PCR-SSP),and its frequency distribution were analyzed and compared between two groups.Results The gene frequency of *1301/1302 allele in the control group was 0.06573.In drug user group,the frequency was 0.04167,RR=0.600649,and decreased but not significantly compared with control group(P>0.05).In HIV infected drug users,the frequency was 0.01220,RR=0.1700,and decreased significantly compared with control group(P<0.05).Compared with drug users group,there was no significant difference in the frequency between drug users and HIV infected drug users(P>0.05).Conclusion There exists close relationship between HLA-DRB1*1301/1302 alleles and resistance of drug use with HIV infection,which plays a crucial role in the pathology,treatment and immune prevention of the disease.
