能获取淋巴结微转移细胞株的鼠结肠癌模型的建立

【摘要】 目的建立一种能获取淋巴结微转移细胞株的结肠癌动物模型。方法将小鼠结肠低分化腺癌Colon26细胞株接种于同源基因鼠BALB/c小鼠爪垫25d后,摘取腘窝淋巴结,用RT-PCR方法检测淋巴结CD44V和CK20基因表达,并将淋巴结粉碎培养于胎牛RPMI1640培养基中观察是否有肿瘤细胞生长。结果腘窝淋巴结可发生淋巴结肿大,而常规淋巴结病理检查不能发现转移灶,但将摘取的肿大淋巴结经RT-PCR检测有CD44V和CK20表达,表达阳性率50%,将其粉碎培养于胎牛RPMI1640培养基中可发现有肿瘤细胞生长,成功率20%,两者比较,差异有统计学意义(P<0.05)。结论运用该动物模型可获得淋巴结中的微转移细胞株,有利于肿瘤微转移的深入研究。

【Abstract】 Objective To establish an animal model of colorectal carcinoma of mice which can acquire micrometastasis cell line in lymph node.Methods The models of micrometastasis in mice were established by injecting murine colonrectal carcinoma cell line-Colon26 into the nail pad of 10 homogenous BALB/c mice.The lymph nodes in popliteal fossa were obtained in asepsis conditions after 25d,and CD44v and CK20 genes in lymph nodes were detected by RT-PCR,then the lymph nodes were granulated to be cultured in RMPI 1640 medium supplemented with 10% fetal calf serum,then,whether there was tumor development was observed.Results CD44v and CK20 genes in lymph nodes were detected in 50% of the mice,and carcinoma cell line could be acquired in lymph nodes in 20% of mice.Conclusion The animal model which can acquire micrometastasis cell line in lymph node of mice is successfully established,which may contribute to the advanced research about tumor micrometastasis.
