大连刺参体腔液甘露糖结合凝集素序列的鉴定及分析

【摘要】 根据NCBI提交的刺参体腔液(Apostichopus japonicus)甘露糖结合凝集素(Mannan-binding lectins,MBLs)序列(AY625513)设计引物,抽取刺参体腔液进行总RNA提取,采用RT-PCR技术,获得498 bp目的片段。通过克隆、测序确定得到刺参MBLs序列。将得到序列进行核苷酸和氨基酸序列比对,鉴定了大连刺参MBLs序列,并为我国不同区域刺参的进一步分类提供了科学的分子水平依据。

【Abstract】 In this study,a pair of primers was designed according to mRNA sequence(AY625513) of mannan-binding C-type lectin(MBL) of the sea cucumber Apostichopus japonicus.Total RNA was extracted from the coelomic fluid of A.japonicus,and 498 bp of MBL(MBL-AJ) gene thereof was obtained by RT-PCR technology.The MBL-AJ sequence was determined through cloning and sequencing.Finally,the MBL-AJ sequence was identificated by nucleotide and amino acid sequence alignment,which provids a scientific basis for molecular taxonomy of A.japonicus in different areas in China.
