七种水蛭组织细胞可溶性蛋白SDS-PAGE电泳分析

【摘要】 采用垂直平板十二烷基硫酸钠-聚丙烯酰胺凝胶电泳法(SDS-PAGE)对我国7种常见水蛭的组织细胞可溶性蛋白成分进行电泳图谱特性分析。结果表明,7种水蛭组织细胞中可溶性蛋白的SDS-PAGE图谱中多数条带相同,但也存在差异。其中日本医蛭(Hirudo nipponia Whitman)蛋白条带最多,达28个,并且吸血蛭类的蛋白条数要明显大于非吸血蛭类,而3种不同地理分布的菲牛蛭(Hirudi-naria manillensis Lesson)间表达蛋白图谱无明显差异。该研究为在分子水平分析我国水蛭不同种类、同种不同地理分布及不同食性间的差异积累了新的生物学数据,对其分类和药材鉴别提供了参考依据。

【Abstract】 The histiocyte soluble proteins of seven leeches in China were analyzed through SDS-PAGE.The results showed that the seven leech species shared most of the soluble proteins components of histiocyte;while a few differences existed.Hirudo nipponia Whitman had the largest number of protein bands of 28.There were more protein bands of blood-sucking leeches than that of non-bloodsucking leeches.No significant difference was detected among the histiocyte soluble proteins components of H.manillensis Lesson from three different areas.It was proved that SDS-PAGE could be used for classification and identification of leeches of different varieties,geographic distribution and food habit at the molecular level,and it accumulated data base of biologicalproperty.
