高效液相色谱法测定人血浆中维生素E的血药浓度
Determination of vitamin E in human plasma by HPLC
【摘要】 目的建立人血浆中维生素E的HPLC测定方法。方法采用维生素K为内标,用甲醇沉淀蛋白后正己烷提取,色谱柱为SymmetryC18(4.6mm×150mm,5μm),流动相为四氢呋喃:甲醇(V/V,18∶82),流速为1.0mL.min-1,柱温40℃,检测波长280nm。结果维生素E在0.625~40mg.L-1范围内线性关系良好,回归方程为y=12.255x-0.6646,r=0.9994,平均绝对回收率74.69%,方法回收率在99%~104%之间。高、中、低3种浓度的日内、日间RSD均小于6%。结论本方法简单、快捷、灵敏、准确,适用于维生素E人体血药浓度的检测。
【Abstract】 OBJECTIVE To establish a method of determining plasma vitamin E in human by HPLC.METHODS vitamin K was used as the internal standard,protein precipitated by methanol and extracted by N-hexane.Separation was carried out on a Symmetry C18(4.6mm×150mm,5μm),the mobile phase consisted of tetrahydrofuran-methanol(V/V,18:82),the flow rate was 1.0mL·min-1,temperature of column was 40℃,UV detection wavelength was 280nm.RESULTS A good linearity was demonstrated between 0.625~40mg·L-1,y =12.255x-0.6646,r=0.9994.The mean absolute recovery was 74.69%,and the method recovery was 99%~104%.The intra-day and inter-day deviation was both showed by RSD<6%.CONCLUSION The method is simple,rapid,sensitive,accurate,and validated for determination of plasma vitamin E in clinical practice.
