微囊化hES/293细胞生物学性状研究

【摘要】 目的检测海藻酸钠-壳聚糖-海藻酸钠(ACA)微囊化人内皮抑素/293(human En-dostatin/293,hES/293)细胞体外培养状况下的生物学性状。方法实验研究分为两组:A组:hES/293细胞未用ACA微囊包裹;B组:hES/293细胞用ACA微囊包裹,每组6个样本。采用聚电解质络合法制作ACA微囊化hES/293细胞,台盼兰染色计数微囊化细胞的存活率和细胞总数,MTT法测定细胞生长状况,流式细胞术检测细胞周期。结果在实验观察的14d内,hES/293细胞可在微囊内以细胞团的形式生长,囊内细胞总数随培养时间的延长而增加,但细胞存活率呈下降趋势;B组细胞在前3 d生长缓慢,第5 d以后生长速率基本上相当于A组;MTT法检测两组在1、3、5、7d的OD值差异无统计学意义(t1=0.208,P1=0.843;t3=0.447,P3=0.674;t5=0.917,P5=0.401;t7=1.956,P7=0.108);流式细胞术显示,B组细胞处于S期的百分比在第7天高于A组,差别有统计学意义(t=3.341,P=0.020),持续时间可达14d,且第14 d二者百分比差异有显著性(t=10.115,P=0.002)。结论微囊化hES/293细胞具有较高的增殖和代谢活性,且能较长时间保持此活性。

【Abstract】 Objective To study the growth and viability of human Endostatin/293(hES/293) cells in the Alginate-chitosan-Alginate(ACA) microcapsules.Methods Two groups were divided which were group A(hES/293 cells non-encapsulated in ACA microcapsule) and group B(hES/293 cells encapsulated in ACA microcapsule).Each group included 6 samples.The hES/293 cells encapsulated in ACA microcapsule were prepared by polyelectrolyte complexometry technology.The biological characters of hES/293 cells were observed by trypan blue staining,the cytoactive and mitotic cycle was detected by methyl thiazolyl tetrazolium(MTT) and flow cytometry respectively.Results hES/293 cells were able to grow and survive as cell clusters in the microcapsules during 14 days.The total cell number per capsule was increasing,but the cell viability rate was decreasing.The cells in group B grew slower than that of group A in the first 3 days,but two groups were similar in the fifth day.The OD-Value of two groups was notstatistic difference in 1,3,5,7 days.The percentage of S phase in group B was higher than group A in the 7th day,and the difference lasted until the 14th day.Conclusion The hES/293 cells encapsulated in ACA microcapsule have not only higher proliferating and metabolic activity,but also keep longer time.
