【摘要】 目的:摸索猪前脂肪细胞的原代培养方法,为研究猪脂肪发生的分子机制奠定基础。方法:取皮下脂肪,酶消化法分离细胞,用转铁蛋白、氢化可的松、胰岛素诱导分化,油红O染色鉴定。结果:在培养期间,细胞逐渐由梭形变为圆形,并逐渐增大,油红O染色为红色。结论:分离的细胞为未分化的前脂肪细胞,可用此方法培养的细胞进行脂肪发育过程中具体分子事件的研究。更多还原

【Abstract】 Objective to establish a primary porcine preadipocyte culture method for better understanding the molecular events of adipogenesis.Methods porcine preadipocyte were iso-lated from subcutaneous adipose using enzemy digeslated method,cultured in 10% fetal bovine serum(FBS)-DMEM/F-12 medium supplemented insulin,transferring and hydrocortisone and identified using Oil Red O.Results cultured fibroblast-like cells were became round during induced differentiation period,the rounded cells were red by Oil Red O staining Conclusion the cultured fibroblast-like cells were preadipocyte.,it could be used to further study for adipose.更多还原
